What is the formula to calculate the Acetylcholine (ACh) concentration using the measured fluorescence intensity of the sample and the standard curve with the Amplex Acetylcholine/Acetlycholinesterase Assay Kit (Cat. No. A12217)?

The fluorescence/absorbance response depends on the sample, microplate, detector settings and other experimental parameters. Therefore, we do not have a general formula to convert fluorescence/absorbance intensity into the concentration of ACh. We recommend generating a standard curve for the assay and fitting it to either a linear, logarithmic or a polynomial curve. Powers 2, 3 or 4 can be used to optimize the best fit for polynomial curves.
