我有两种来自于同一宿主种属的一抗,我希望用它们来标记同一种细胞,该如何选择二抗?

如果这两种一抗来自不同亚型(如小鼠IgG1和IgG2a),则可选择该亚型特异性的二抗。另一种办法是通过抗体标记试剂盒、活性染料或定制标记,直接与一抗进行偶联反应。

What storage conditions would you recommend for secondary antibodies conjugated with fluorophores or HRP?

We recommend storing secondary antibodies conjugated with fluorophores or HRP at 4 degrees C as we have found that storage at -20 degrees C and the associated continuous freeze/thaw cycles can reduce the performance of fluorescent dyes. Storage at 4 degrees C improves fluorophore stability without affecting antibody performance.
You can also prepare small aliquots to avoid freeze/thaw cycles and store them at -20 degrees C, as with each freeze/thaw step the performance of the antibody would go down.

I have two primary antibodies from the same host species that I wish to use to label the same cells. How do I choose secondary antibodies?

If the two primary antibodies are from different isotypes (such as mouse IgG1 and IgG2a), there are isotype-specific secondary antibodies to choose from. The other option is to make direct conjugates of the primary antibodies through the use of antibody label kits, reactive dyes, or custom conjugations.
