What is the advantage of using phospho-enrichable crosslinkers?

Phospho-enrichable crosslinkers such as DSPP (Disuccinimidyl Phenyl Phosphonic Acid, PhoX) and TBDSPP (tert-Butyl Disuccinimidyl Phenyl Phosphonate, tBu-PhoX) are great tools for studying protein-protein Interaction, protein 3D structure, and for mass spectrometry (MS) analysis. They can be used to enrich low-abundance crosslinked peptides and thereby improve MS identification rates.

Yes, TBDSPP is cell-permeable. After the crosslinking reaction and before the phospho-enrichment step, TBDSPP needs to be de-protected.
