How large is the sample imaging area on the iBright Imaging Systems?

The imaging area of the iBright Imaging system is 22.5 x 18.0 cm. This gives ample space for imaging 4 mini blots (or 2 midi blots) simultaneously.

What sample types can I image with the iBright CL750 Imaging System?

The iBright CL750 Imaging System supports imaging of chemiluminescent western blots, colorimetric and fluorescent stained gels, and nucleic acid gels. It can also be used to image GFP- and crystal violet-stained colony plates.

What sample types can I image with the iBright CL1500 Imaging System?

The iBright CL1500 Imaging System supports a wide range of applications including the following:

Protein gel: colorimetric staining of gels (e.g., Coomassie, silver) and membranes (e.g., Ponceau S, Thermo Scientific Pierce Reversible Protein Stain), fluorescent staining of gels (e.g., Invitrogen SYPRO Ruby stain)

Nucleic acid gel: ethidium bromide and Invitrogen SYBR stains; imaging of E-Gels and hand pour gels is supported

Chemiluminescent western blot: chemiluminescence using all popular HRP and AP substrates (e.g., Thermo Scientific SuperSignal and Invitrogen WesternBreeze substrates)

Additional applications: colony counting for GFP, crystal violet, and blue/white colonies; TLC plates, and leaf sections

What sample types can I image with the iBright FL1500 Imaging System?

The iBright FL1500 Imaging System provides maximum application support including up to 4-channel multiplex western blot imaging. See the list below for more details:

Protein gel: colorimetric staining of gels (e.g., Coomassie, silver) and membranes (e.g., Ponceau S, Thermo Scientific Pierce Reversible Protein Stain), fluorescent staining of gels (e.g., Invitrogen SYPRO Ruby stain)

Nucleic acid gel: ethidium bromide and Invitrogen SYBR stains; imaging of E-Gels and hand-poured gels is supported

Chemiluminescent western blot: chemiluminescence using all popular HRP and AP substrates (e.g., Thermo Scientific SuperSignal and Invitrogen WesternBreeze substrates)

Fluorescent western blot: fluorescence with popular RGB (visible range) and near-IR fluorophores (e.g., Invitrogen Alexa Fluor and Alexa Fluor Plus conjugates)

Which iBright imager models are compatible with the iBright System Contrast Tray (Cat. No. A56597)?

All iBright imager models are compatible with the orange iBright System Contrast Tray (Cat. No. A56597), inclusive of the CL1000, FL1000, CL750, CL1500, and FL1500 iBright imager models. What sample types can I image with the iBright System Contrast Tray (Cat. No. A56597)?

We recommend using the iBright System Contrast Tray (Cat. No. A56597) for imaging all visible protein gel samples such as those stained with Coomassie Blue or Silver Stain. For more information on compatible reagents and applications, visit the iBright Imaging Systems Compatible Reagents page.

What is the best way to image Ponceau staining with an iBright Imaging System?

The recommended imaging method depends on the iBright imager model being used. For iBright FL1500 and CL1500 Imaging Systems, we recommend imaging Ponceau-stained samples with the Ponceau application that can be found in Universal mode. For the iBright CL750 Imaging System, we recommend imaging Ponceau-stained samples in Protein gel mode.

We always recommend imaging with the orange iBright System Contrast Tray (Cat. No. A56597), and if needed, adjust the focus or zoom to get bands as sharp as possible. Better quality pictures are obtained when the membrane is wet.

Can I use the No-Stain Protein Labeling Reagent with an orange or red dye (i.e., will the signal appear in channel 2, 3, 4 or 5, not just channel 1 in the iBright imager)?

Channels 3, 4 and 5 on the iBright imager can be used for multiplexing without interference while using the No-Stain Protein Labeling Reagent. Channels 1 and 2 on the iBright imager will show interference. Fluorophores for multiplexing that are compatible with the No-Stain Protein Labeling Reagent are listed here, under the “Secondary Abs” tab (https://www.thermofisher.com/us/en/home/life-science/protein-biology/protein-assays-analysis/western-blotting/detect-proteins-western-blot/total-protein-normalization.html).
