The CYQUANT LDH Cytotoxicity Assay Kit (Cat. Nos. C20300, C20301) Assay Buffer used to prepare the Reaction Mixture, contains the reaction dye and therefore has some inherent coloration.
The color change should be considerably noticeable with the Positive Control in comparison to the Assay Buffer alone.

We do not recommend doing this. The LDH in the media should be tested as soon as possible to avoid the loss of LDH activity. The reaction mixture is stable for 3 to 4 weeks, stored frozen.

We do not recommend doing this. Refrigeration or freezing and storage over time would cause the loss of LDH activity.

Serum may contain endogenous LDH that can interfere with the assay. To determine feasibility, we recommend testing a cell-free sample of media and serum only, using the same lot of serum to be used on your samples. If different lots of serum are to be used, each lot must be tested for endogenous LDH.
