DAF-FM 是用于检测和定量低浓度一氧化氮 (NO) 的试剂。DAF-FM 本身不发荧光,但与 NO 反应后形成荧光产物苯并三唑。能检测荧光素的仪器均可以检测 DAF-FM 荧光,包括流式细胞仪、显微镜、荧光酶标仪和荧光计。

DAF-FM 质量标准:
•DAF-FM Ex/EM:∼495/515 nm
• 冻干品应使用 DMSO 溶解,然后用水性缓冲液稀释,制成工作溶液
• DAF-FM 二乙酸酯具有细胞可透过性并且能够被动穿透细胞膜;一旦进入细胞,它就会转换为细胞不可透过形式
• 含牛血清白蛋白 (BSA) 和酚红的缓冲液可能会影响荧光,应谨慎使用
• DAF-FM 的荧光量子产率为 ∼0.005,但与 NO 反应后产率达到 ∼0.81 左右,约增加了 160 倍

• 通过流式细胞分析评估转醛醇酶缺陷淋巴母细胞中 NO 的产生
• 检测神经营养因子刺激后胚胎皮层神经元中 NO 的蓄积
• 斑马鱼中 NO 的体内成像

DAF-FM 相对于 DAF-2 的明显优势
DAF-FM 的 NO 加合物光谱与 pH 值高于 5.5 无关。此外,DAF-FM 的 NO 加合物的光稳定性显著高于 DAF-2,这意味着需要延长图像采集时间。总之,DAF-FM 试剂对 NO 的灵敏度高于 DAF-2(对于 DAF-FM,NO 的检测限 ∼3 nM;对于 DAF-2,NO 的检测限 ∼5 nM)。
