Range of separation in 1X TAE: 0.6%: 20,000-1000 bp
0.8%: 12,000-500 bp
1.0%: 8000-300 bp
1.2%: 6000-200 bp
1.5%: 3500-100 bp
2.0%: 2000-50 bp

How do I prepare an agarose gel?

We recommend the following to prepare an agarose gel:

  1. Weigh out the desired amount of agarose and place it in an Erlenmeyer flask with a measured amount of electrophoresis buffer. Make sure to use the same electrophoresis buffer in the gel as for the running buffer.
  2. Example: For a 0.8% gel, add 0.8 g of agarose and 100 mL of TBE Buffer (1X), to a 200 mL flask. The larger flask insures against the agarose boiling over.
  3. Dissolve the agarose in a boiling water bath or in a revolving-plate microwave oven. All the grains of agarose should be dissolved, and the solution should be clear.
  4. Cool the solution to 60°C (70°C for concentrations 2% or above) and pour immediately.
  5. Allow the gel to set for 30 minutes before using.
