We recommend to evaluate purity with NanoDrop and measure quantity of dsDNA with Qubit$reg; before NGS library preparation. Results can vary depending on the FFPE samples used.
Due to high de-crosslinking efficiency of GeneJET FFPE DNA Purification Kit, high-quality purified DNA can be directly used in sensitive downstream applications such as qPCR, PCR (amplicons up to 1 kb) or stored at -20 degrees C.
Paraffin sections can be stored at or below +4 degrees C for 1 year without observable negative effects on DNA yield and usability. Longer-term storage of FFPE sections may have negative effects on the DNA due to oxidation.
DNA yield and quality from FFPE tissue may vary considerably depending on the tissue source, the thickness of the slice, how much tissue is in the slice, the age of the sample, post-sampling delay before fixation, fixation time, etc. We recover up to 8 µg of genomic DNA from one slice (10 µm thickness) of FFPE sample depending on the tissue type.
Up to 8 sections (up to 10 µm thickness) of FFPE tissue samples can be used for DNA isolation.
We have not tested the GeneJET FFPE DNA Purification Kit with fixed (non paraffin-embedded) tissue.
