Nedisertib, also known as M3814, is a potent and selective inhibitor of DNA-dependent protein kinase (DNA-PK), currently under phase 2 clinical trials. This kinase plays a crucial role in the non-homologous end joining pathway of DNA repair, making nedisertib a promising candidate for cancer therapy, particularly in combination with radiotherapy and chemotherapy agents like capecitabine. The drug's mechanism involves modulating ABCG2-mediated multidrug resistance, which is a significant challenge in cancer treatment. Nedisertib's ability to inhibit DNA-PK enhances the efficacy of radiotherapy by preventing the repair of radiation-induced DNA damage in tumor cells, thereby promoting cell death.
