Amantadine is a well-established antiviral and antiparkinsonian agent, primarily targeting the NMDA receptor to exert its therapeutic effects. It is located on chromosome 1 at the locus 1p36.23, and its chemical structure is characterized by a tricyclic amine with a cage-like adamantane core, which contributes to its unique pharmacokinetic properties. Functionally, amantadine blocks viral replication by interfering with the viral M2 protein, and in Parkinson's disease, it modulates dopaminergic transmission, providing symptomatic relief. Recent studies have explored its hybridization with gardenamide A to enhance NMDA antagonism, indicating potential for expanded therapeutic applications.
