Can I use SILAC if I do not have access to an AB/MDS instrument?

Yes, SILAC reagents can be used with other instruments, but you need to carry out the quantitation manually or ask the instrument manufacturer for analysis software packages for automated protein quantitation. Some manufacturers may have new software packages for this application.

Does SILAC work using LC/MS?

Yes. LC/MS can be used to analyze samples labeled by SILAC. Always consult with your mass spectrometry facility to determine what protocols are most compatible with their instruments.

Can I use 2-D gels to analyze proteins from cells labeled by SILAC?

Yes. Samples labeled by SILAC can be analyzed by 2-D or in conjunction with 1-D gels to validate results, analyze difficult proteins, and screen for specific proteins that are hard to identify by 1-D SDS-PAGE.

Are the heavy amino acids included in the price of the SILAC kits?

Yes, the kit includes all the components for labeling cells with the heavy lysine. Heavy arginine is sold as a separate item (Cat. No. MS10009) for researchers who want to perform a double-label experiment using heavy lysine and arginine.
