Western blot was performed using Anti-DNA Ligase I Polyclonal Antibody (Product # PA1-29815) and a 130kDa band corresponding to DNA ligase 1 was observed across cell lines tested. Nuclear enriched extracts (30 µg lysate) of HeLa (Lane 1), Raji (Lane 2), A-431 (Lane 3), A549 (Lane 4), HT-29 (Lane 5), MIA PaCa-2 (Lane 6), MCF7 (Lane 7) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). This gene complements a mutation in the temperature-sensitive hamster mutant ts11, which blocks progression through the G1 phase of the cell cycle at nonpermissive temperature. There are three alternatively spliced transcript variants encoding the same protein described for this gene.
