Immunocytochemical analysis of SENP8 in Hela cells using a SENP8 polyclonal antibody (Product # PA5-118500). The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with the primary antibody (1:200) and mouse anti-beta tubulin antibody at a dilution of 1:200 for 1 hour at 37°C. SENP8 (Cysteine protease FKSG8; DEN1; FKSG8; HsT17512; NEDP1; PRSC2; SUMO/sentrin specific protease family member 8), is a novel cysteine protease which belongs to the ULP family of deubiquitinases. ULP family members consist of a C-terminal core catalytic domain and N-terminal extensions determining substrate specificity. SENP8, a 24KDa protein, plays a major role in regulating NEDD8 pathway. It processes preNEDD8 to its mature form and deconjugates NEDD8 from substrates such as Cullins and p53. SENP8 may play a role in preventing accumulation of hyper-neddylated cullin and maintaining the level of mono-neddylated cullin that is thought to increase the efficiency of ubiquitination by SCF ubiquitin ligases.
