Antibody detects endogenous levels of fragment of activated Caspase 8 resulting from cleavage adjacent to Asp384.
Caspase-8 (cleaved Asp384) refers to the activated form of caspase-8 generated following proteolytic cleavage at aspartic acid residue 384, a key step in the activation of this initiator caspase in the extrinsic apoptosis pathway. Caspase-8 is encoded by the CASP8 gene and is synthesized as an inactive zymogen (pro-caspase-8) containing tandem death effector domains (DEDs) that enable recruitment to death-inducing signaling complexes (DISCs) formed downstream of death receptors such as Fas (CD95), TRAIL receptors, and TNF receptor 1. Upon DISC assembly, pro-caspase-8 undergoes dimerization and autocatalytic cleavage at specific aspartic acid sites, including Asp384, producing active subunits that assemble into the functional enzyme. Activated caspase-8 initiates the apoptotic cascade by cleaving and activating downstream effector caspases such as caspase-3 and caspase-7, and can also process BID to link extrinsic and intrinsic apoptosis pathways. Detection of cleaved caspase-8 (Asp384) is widely used as a marker of death receptor-mediated apoptotic signaling in cell death and cancer research.
