You cannot use Anti-His (C-term) antibody against the N-terminal HisG epitope taq. Thermo Fisher Scientific sells two Anti-His antibodies: Anti-HisG and Anti-His (C-term) antibodies depending on the epitope tag in your vector. These antibodies do not recognize react well with both epitopes because the glycine and the carboxy terminus are important parts of each one. Any 6xHis tag lacking in these additional elements is a poor epitope for either of the antibodies.

No, the Anti-HisG antibody detects the N-terminal polyhistidine (6x His) tag followed by glycine: HHHHHHG. The proteins made from the pDEST17 vectors do not have this glycine following the His tag and therefore will not be recognized by this antibody.
