SiteClick Duet and Quartet antibody labeling kits are used to create label-ready, site-specific, azido-modified antibodies without lengthy genetic modifications. Our enzyme-driven SiteClick labeling precisely attaches an azido moiety to the heavy chains of IgG antibodies, preserving antigen-binding domains for optimal target binding. This site selectivity targets carbohydrate domains on the Fc region, applicable to all IgG antibodies regardless of isotype or host species.

The SiteClick Quartet Antibody Labeling Kit offers up to four payloads per antibody (DOS 4) in less than 24 hours, while the SiteClick Duet Antibody Labeling Kit enables the attachment of two payloads (DOS 2) in only 4 hours.

Features of the SiteClick Duet and Quartet antibody labeling kits include:

  • Modify any IgG antibody species or isotype with a complete kit
  • Follow a brief, straightforward protocol
  • Preserve antibody function with efficient, site-specific labeling
  • Achieve consistent results with reproducible labeling chemistry
  • Develop therapeutic antibodies, antibody-drug conjugates, antibody-oligo conjugates, bispecific antibodies, and immunodiagnostics

Once azido-modified, choose from a variety of SiteClick sDIBO or DBCO alkyne labels for covalent attachment via copper-free click chemistry under mild conditions. Enjoy the flexibility to select different fluorescent labels, including Alexa Fluor, pHrodo, and LysoLight dyes, or to conjugate other molecules such as biotin, oligos, toxins, or degraders.

SiteClick labeling is useful for developing therapeutic antibodies, antibody-drug conjugates, biologics, and immunodiagnostic tests, helping ensure high specificity, reproducible labeling, and preserved antibody performance.
