No, the presence of BSA will not influence the labeling efficiency of the SiteClick Kit. Glycoproteins at a high concentration may affect the labeling efficiency and since BSA is not a glycoprotein, it does not inhibit labeling efficiency with the UDP-GalNAz component of the SiteClick kit.

Yes. We highly recommend using an sDIBO alkyne in order to limit non-specific binding to the antibody, but the azide would react with any alkyne (using copper-mediated click reaction) or any DIBO or sDIBO alkyne (using a copper-free click reaction).

We do not recommend doing this. Although the sDIBO alkynes will react with any azide-modified molecule, SiteClick Antibody Azido Modification Kit was optimized specifically for antibody labeling.
