SYBR Safe DNA 凝胶染色剂专门开发用于降低致突变性–,比用溴化乙锭在琼脂糖或丙烯酰胺凝胶中对 DNA 进行染色更安全。SYBR Safe 染料不仅比溴化乙锭具有更低的致突变性,而且 SYBR Safe 染料的检测灵敏度比溴化乙锭更高。SYBR Safe 染料作为一种预混合溶液,可以像溴化乙锭溶液一样使用,既可以在电泳过程中在凝胶中使用,也可以作为电泳后的染料使用。与核酸结合时,绿色荧光 SYBR Safe 染料在 ∼280 和 ∼502 nm 处具有荧光最大激发波长,在 ∼530 nm 处具有荧光最大发射波长。SYBR Safe DNA 凝胶染色也可作为 1L (S- 33100) 即用型溶液提供。SYBR Safe DNA 凝胶染色剂入门试剂盒 (S-33110) 包括 1 L SYBR Safe DNA 凝胶染色和一个拍照滤光片 (S-37100)。

使用SYBR Safe DNA凝胶染料染色,为什么我有时候会在凝胶内看到斑点?

很多衣服上的增白剂,以及某些真菌和细菌,和SYBR Safe DNA凝胶染料会发出同样波长的荧光。这些位于凝胶表面或凝胶内的污染物可能会产生斑点。

SYBR染料的pH范围是多少?

SYBR染料仅在很窄的pH范围内有效,约为pH 7-8。超出此范围,荧光信号将迅速消失。

电泳时,SYBR Safe染料朝哪个方向迁移?

与溴化乙锭相似,SYBR Safe DNA凝胶染料与DNA的迁移方向相反。由于只有凝胶的最底部染料浓度会在电泳过程中降低,所以使用SYBR Safe DNA凝胶染料制备的凝胶没有实际影响。在电泳结束后用SYBR Safe DNA凝胶染料染胶,可部分抵消这种作用。染料溶液不应该加入到电泳缓冲液中,因为这可能会导致染料在电极上分解,并将有毒的挥发性化合物释放到空气中。
