Insulin-Transferrin-Selenium-Ethanolamine (ITS-X) Select Supplement is an animal-origin-free (AOF) basal medium supplement that is used to reduce the amount of FBS needed to culture cells.

Gibco Insulin-Transferrin-Selenium-Ethanolamine (ITS-X) Select Supplement is an animal-origin-free (AOF) basal medium supplement. The insulin and transferrin in the ITS-X Select formulation are produced in non-mammalian recombinant expression systems.

ITS-X is used as a basal medium supplement to reduce the amount of fetal bovine serum (FBS) used to culture cells. It is routinely used, in combination with 2–4% FBS, to support cell types that require ethanolamine, such as bronchial epithelial cells.

Insulin promotes glucose and amino acid uptake, lipogenesis, intracellular transport, and the synthesis of proteins and nucleic acids. Transferrin is an iron carrier and may also help reduce toxic levels of oxygen radicals and peroxide. Selenium, as sodium selenite, is a co-factor for glutathione peroxidase and other proteins that is used as an antioxidant in media. Ethanolamine is a precursor of phospho-glycerides, which are essential to the structure of the plasma membrane and cellular organelles.

The product formulation contains the following components:

  • Insulin (1000.0 mg/mL)
  • Transferrin (550.0 mg/mL)
  • Sodium selenite (0.67 mg/mL)
  • Ethanolamine (200.0 mg/mL)

What is the difference between Insulin-Transferrin-Selenium (ITS) (100X) and Insulin-Transferrin-Selenium (ITS) Select (100X) supplements?

ITS Select supplements are animal origin-free (AOF) versions of the ITS supplements offered by Thermo Fisher Scientific.

ITS Select supplements use recombinant versions of both proteins in the formulation: insulin and transferrin.

All other versions of ITS, those without the Select designation, utilize recombinant insulin and a human-derived holo transferrin.

All ITS and ITS Select supplements have the following concentrations of their components:

Transferrin: 0.55 g/L

Can I store aliquots of Insulin-Transferrin-Selenium (ITS) (100X) supplements at -20 degrees C without compromising the integrity of the product?

We recommend storing all ITS and ITS Select supplements at 2-8 degrees C as there is a chance that the insulin will precipitate upon freezing and thawing.
