EnzChek™ 磷脂酶 C 直接检测试剂盒为监测 PC-PLC 活性提供了一种简单可靠的方法。每个试剂盒都能提供足够2块96孔微孔板以 200 µL 反应体积进行检测的试剂。PC-PLC 在涉及细胞凋亡和细胞存活的多种细胞信号转导通路以及从癌症到 HIV1-7 的各种疾病中发挥着至关重要的作用。该测定试剂盒使用带有染料标记 sn-2 酰基链的甘油磷酸乙醇胺作为 PC-PLC 的底物。PC-PLC 在磷酸基之前对底物进行切割并释放出染料标记的二酰甘油,后者产生可连续测量的阳性荧光信号。反应产物的最大吸收和荧光发射波长分别为 509 nm 和 516 nm。采用来自蜡样芽孢杆菌的纯化酶,该测定试剂盒能够在室温条件下在一小时后检测到低至 10 ⁄mU/mL 的 PC-PLC 含量。该试剂盒已经过验证可用于表征 PC-PLC 抑制,因为它是一种直接测量方法,所以消除了在化合物筛选中引起假阳性的可能性。
