The Total Oxidant Status (TOS) Colorimetric Assay Kit is a highly reliable and sensitive assay designed for the quantitative measurement of TOS in cells, tissue, serum, and other liquid sample types. Oxidant molecules, also known as reactive oxygen species (ROS), are produced during normal metabolic processes and can be further stimulated by internal and external factors. When the balance between oxidation and antioxidant defense shifts towards oxidation, it can lead to oxidative stress and various diseases. Measuring the concentrations of different oxidant molecules individually can be time-consuming, costly, and technically complex. Therefore, the Total Oxidant Status (TOS) assay provides a practical solution by quantifying the overall oxidation state of a sample.

In this assay, oxidizing agents in the sample oxidize ferrous ions (Fe2+) to ferric ions (Fe3+), which strongly bind to xylenol orange to produce a blue-purple complex with a maximum absorption wavelength of around 590 nm. The color depth is proportional to the content of oxidation substances in a certain concentration and a certain time and can be used to indirectly calculate the total oxidation state of the sample.
