The BUB1 gene encodes a serine/threonine-protein kinase that is integral to the spindle assembly checkpoint, a crucial mechanism ensuring proper chromosome alignment and segregation during mitosis. BUB1 is involved in detecting and correcting errors in kinetochore-microtubule attachments, which is vital for maintaining genomic stability. It acts by delaying the onset of anaphase until all chromosomes are correctly attached to the spindle apparatus. BUB1 is recruited to kinetochores in early mitosis, where it phosphorylates several substrates to activate other checkpoint proteins, such as BUBR1 and MAD2, thereby preventing premature progression through mitosis. Mutations or aberrations in the expression levels of BUB1 are often associated with aneuploidy and chromosomal instability, contributing to cancer progression. BUB1 is thus considered a potential target for cancer therapeutics, aiming to exploit its role in the mitotic checkpoint to selectively eliminate tumor cells through checkpoint bypass or mitotic arrest.
