Antibody Assays for HLA, Episode 3



Episode 3 of the 2025 Back to Basics series provides a comprehensive overview of bead-based solid phase assays used in the detection and definition of HLA-specific antibodies. A very brief history is given of the evolution of HLA-specific antibody detection assays from the traditional cell-based complement-dependent cytotoxicity test to the bead-based methods that were first introduced in the 1990s with the FlowPRA test. The fundamental principles of the bead-based assays are discussed and include the FlowDSA crossmatch test, FlowPRA and LABScreen antibody screening and identification assays. The episode also covers general protocol key points and troubleshooting tips and a comparison of sensitivity between the different LABScreen assays.
