HLA fundamentals

This comprehensive webinar series, led by Linda Olsen from our technical training team, will provide an in-depth understanding some of the crucial aspects of transplant immunology and genetics.  It aims to support both newcomers and experienced professionals in refreshing their understanding of HLA foundational principles.

Episode 1: Molecular Biology and Genetics

This episode briefly summarizes fundamental knowledge supporting the study of HLA. This will include genetic inheritance, gene expression, and population dynamics, and how genetic information flows from DNA to proteins.
 

Episode 2: Immunology

This session will focus on the processes and components of the innate and adaptive arms of the immune system, including cells, receptors, and pathways important to the understanding of HLA.
 

Episode 3: HLA Proteins and Genes

This episode focuses directly on HLA proteins and genes. It will cover the HLA system in general: the various classes and loci of HLA, their expression and roles within the human body, and specifics of HLA genetics and nomenclature.

 

Episode 4: Anti-HLA Antibodies and Immune Responses

This episode describes antibodies and possible immune responses against HLA proteins. It will cover information about antibodies in general, specific aspects of anti-HLA antibodies, and the possible immune responses in solid organ and stem cell transplant.

Episode 5: Starting with Serology: History and Evolution of HLA Assays

This episode will summarize the development of HLA assays over time, from leukoagglutination, mixed lymphocyte reactions, and complement-dependent cytotoxicity to the more sensitive and specific assays in use today for genetic and immunologic testing related to HLA.
 

Episode 6: Clinical Applications of HLA Testing

This episode describes some of the ways HLA testing is used in clinical and research laboratories. Learners will take away a general overview of HLA testing in transplantation and other applications.

HLA testing methodologies

This webinar series initiative focuses on the scientific principles and technical details of laboratory assays crucial for HLA and transplantation. It aims to support both newcomers and experienced professionals in refreshing their understanding of relevant testing technologies. Each session covers different aspects of HLA testing methodologies and are tailored to provide a deep dive into the techniques, processes, and instrumentation used in HLA labs, making it a comprehensive educational resource.

Episode 1: DNA Assays for HLA: SSP and RT-PCR

DNA Assays for HLA - SSP and RT-PCR covers the principles and practices of SSP and RT-PCR techniques in HLA typing. It details workflows including PCR, gel electrophoresis, and LinkSeq for as DNA methods for HLA laboratories. The presentation underscores the continued importance of these classic DNA methods in HLA laboratories for their efficiency and rapid results.

Episode 2: DNA Assays for HLA: SSO

This presentation details DNA assays for HLA using the SSO method, focusing on LABType protocols. It includes steps for amplification, hybridization, washing, and instrument analysis. Key topics include intended results, data collection, reporting, and laboratory applications, ensuring comprehensive coverage of SSO practices and principles.

Episode 3: Antibody Assays for HLA

The “Bead-based antibody detection assays” episode covers the evolution and fundamental principles of antibody detection assays for HLA-specific antibody screening and identification. Flow cytometer bead based tests include the FlowDSA crossmatch test and FlowPRA antibody screening and identification assays, whilst the Luminex bead based tests include LABScreen antibody screening and identification assays. General protocol key points, troubleshooting tips and sensitivity differences between the tests are discussed.

Episode 4: Flow Cytometry for HLA

This presentation introduces flow cytometry with a focus on HLA-related clinical applications. It explains the basic operation of a flow cytometer, immunophenotyping to identify cell populations, and outlines the flow cytometric crossmatch (FCXM) in transplantation, along with an overview of additional HLA flow technologies used for antibody detection and crossmatch refinement.

Episode 5: Principles of NGS

In the final installment of our series, we will introduce some of the critical terms used in NGS assays and the differences in methodologies and instrumentation to provide a large-scare view of the basic principles of NGS.  Topics covered will be the Utility of NGS typing, NGS Key Terms, Methodologies and instrumentation.
