Simultaneous Quantitation and Discovery (SQUAD)

For fully parallelized acquisition of both targeted and untargeted data, Orbitrap Tribrid mass spectrometers utilize both Orbitrap and ion trap mass analyzers simultaneously. During SQUAD analysis on Orbitrap Tribrid Apex Small Molecule MS, full scan MS1 profiling is performed in the Orbitrap for untargeted discovery data acquisition while known metabolites of interest are detected using targeted-MS2 experiments with sensitive ion-trap detection. The improved ion-funnel and increased ion flux effect overall sensitivity when compared to previous generation small molecule Tribrids.

Compared to full scan Orbitrap quantitation, targeted MS2 ion-trap methods often exhibit an additional order of magnitude linearity at the low end of the curve achieving a linear dynamic range of seven orders of magnitude.

 

The increased sensitivity of Orbitrap Tribrid Apex Small Molecule MS provides up to 10x improvement over the previous small molecule generation Orbitrap Tribrid MS.


Reveal structural complexity with MSn and UVPD

Lipids have a high capacity for isomers due to the positions of their double bonds or even a simple change in position for the acyl chains. Elucidation of the isomeric species requires in-depth mass spectrometry. The ion trap allows for MSn experiments with alternate fragmentation, like UV-laser based dissociation. By utilizing pre-built templates and multiple tools available on the system, distinguishing small molecule isomers is made possible.

Two isomeric lipids differ only in the position of the acyl chains. UVPD MS3 produces distinct fragments that distinguish isomers for confident structural annotation.


FAQs

There are significant hardware and software changes between the previous generation Orbitrap Tribrid MS and the Orbitrap Tribrid Apex Small Molecule MS. Improvements include the addition of a brighter source with the letter-box-shaped ion transfer tube and a front ion routing multiple, which improves the parallelization of ion transfer and increases the number of scans possible in the same timeframe. IR-laser for alternate fragmentation capabilities has also been added.

The Small Molecule edition is the most unique edition within the Orbitrap Tribrid Apex MS line. It is tailored for the specific needs of the small molecule community without the extra capabilities needed for proteomics or protein structural analysis. This means that certain features, including TurboTMT, data-independent acquisition, and data-dependent acquisition for charge state above +2 are not available. The mass range is limited to lower m/z values. This allows researchers to focus on their application space without features they will not use. However, if you become interested in expanding your capabilities in the future, we offer field upgradeability to the MultiOmics Edition, to ensure your future needs are met.

The UV-laser option is recommended for the Orbitrap Tribrid Apex Small Molecule MS.

Yes, but the roughly 30% increase in MS1 fragment for Phenylalanine is also accompanied by a 10x improvement in sensitivity and another order of magnitude linear range due to the new letter-box opening ion transfer tube. The Orbitrap IQ-X Tribrid MS has the round bore opening ion transfer tube, which helps reduce source fragmentation, but also reduces ion transfer into the system.
