Thymine is normally found as a deoxyribose. Thus, TTP and dTTP are essentially the same thing. Both types of mixes can be used in PCR reactions
Uracil-N-glycosylase (UNG) has activity in the temperature range of 4 degrees C to 55 degrees C, and is not completely inactivated by incubation at 95 degrees C. It is recommended that the annealing temperature be at or above 55 degrees C when using this enzyme with any of our PCR or Sequence Detection Systems kits to avoid possible degradation of newly synthesized dU-containing PCR products by residual UNG activity. For this reason, it is also recommended that the post-PCR soak be maintained at 72 degrees C (instead of 4 degrees C) if analysis of the PCR product will be performed using a gel-based method.

The four dNTPs (dATP, dCTP, dTTP, dGTP) are used at a final concentration of 200 µM each or a total of 0.8 mM in standard amplifications. For PCR amplifications that require high fidelity, lower dNTP concentrations (on the order of 10-50 µM) work optimally.

The control primers generate a 300 and a 500 bp amplicon from the lambda control DNA. The Lambda Multiplex Control primers, as well as the Lambda Control DNA are provided in this kit.
