Can the Dye Removal Columns be reused?

No. The resin is effective for one-time use only and cannot be regenerated.

Yes, the resin is compatible with up to 10% organic solvent such as DMF.

Which fluorescent dyes can I remove with the Dye Removal resin?

DyLight Dyes, rhodamine and fluorescein have been removed successfully using this resin. Samples with dyes in the green spectrum, such as Dylight 488 Dyes, might require double processing because they require a high molar excesses for labeling. The ratio of resin to protein/dye mixture is typically 1:1. When using fluorescent dyes other than those indicated, the ratio of resin to protein/dye mixture will require optimization.

How many samples can I process with the Dye Removal Columns?

The kit includes sufficient material to process 10 samples of 250 µl each for most dyes. The kit contains 5 mL of Dye Removal Resin, 10 Spin Columns and 20 Collection Tubes. The ideal labeled-protein concentration range is 1-2.5 mg/mL.

Can I use the Dye Removal Columns for desalting?

No. The resin is not appropriate for desalting applications.

Will the Dye Removal Columns effectively remove dye from a sample that contains a small protein?

Yes. For best results, use a protein solution at neutral pH (6.5-8.5) with at least 100 mM NaCl to enhance protein recovery. The optimal buffer for purifying proteins < 20 kDa is phosphate-buffered saline (PBS; 100 mM sodium phosphate, 150 mM NaCl; pH 7.2).
