The most commonly used buffer is RIPA Buffer with SDS. We offer RIPA Buffer (Cat. Nos. 89900 and 89901). We also offer the Pierce IP Lysis buffer (Cat. Nos. 87787 and 87788) as well as M-PER (Cat. Nos. 78501, 78503, and 78505).

Pierce IP Lysis Buffer ships at ambient temperature and can be stored at room temperature. However, we recommend 4 degrees C for long term storage in order to prevent possible bacterial growth.

The Thermo Scientific IP Lysis Buffer is compatible with Thermo Scientific BCA Protein Assay (Cat. No. 23225) and Thermo Scientific 660 nm Protein Assay (Cat. No. 22660). It is not compatible with Thermo Scientific Coomassie Plus (Bradford) Assay (Cat. No. 23236) unless diluted 1:1 in TBS, PBS, or water.

For suspension cells, use 500 µl of lysis buffer per 50 mg of wet cell pellet (10:1 v/w). Incubate lysate on ice for 5 minutes with periodic mixing. Remove cell debris by centrifugation at ~13,000 × g for 10 minutes at 4 degrees C. Transfer supernatant to a new tube.

The Thermo Scientific IP Lysis Buffer does not contain protease or phosphatase inhibitors. We recommend inhibitors, such as Thermo Scientific Halt Protease Inhibitor Cocktail (Cat. No. 87786) or Phosphatase Inhibitor Cocktail (Cat. No. 78420) to be added to the buffer just before use.

The Thermo Scientific IP Lysis Buffer is recommended for scientists working with mammalian cells in order to extract cellular proteins for Immunoprecipitation and Western Blot analysis. It is effective for lysing cultured mammalian cells from both plated cells and cells pelleted from suspension cultures, and is especially formulated for pull-down and immunoprecipitation assays, and as a wash buffer for affinity resins. This lysis buffer is also compatible with protein purification, immunoassays and ELISA.

The Thermo Scientific IP Lysis Buffer contains 25 mM Tris.HCl pH 7.4, 150 mM NaCl, 1 mM EDTA, 1% NP-40 and 5% glycerol. This formulation is optimized for compatibility with immunoprecipitation and pull-down assays and does not contain SDS.

The Thermo Scientific IP Lysis Buffer (Cat. No. 87787) is a mammalian whole cell lysis buffer based on a modified RIPA buffer formulation without SDS. This moderate strength lysis buffer is able to effectively solubilize cellular proteins but does not liberate genomic DNA or disrupt protein complexes like RIPA buffer.
