It is possible that there is a communication problem between the Ion GeneStudio S5 Series system and the server. We recommend shutting down the system, waiting for 2 minutes and then rebooting. If that does not work and if you are using the Ion S5 XL System, we recommend turning off the server and the system for 2 minutes. Then, attach a keyboard and monitor to the server and turn the server back on. Log in to the server and make sure you can connect to it via your web browser. Then, turn on the system. If the planned run does not appear, please contact Technical Support at techsupport@thermofisher.com for further assistance.

No. The recommended input amount is 20 ng of cfDNA but successful libraries have been made from 1-50 ng of cfDNA.
We do not recommend using spectrophotometer methods for quantifying cfDNA because they are not reliable with low concentrations of nucleic acids. Use of these methods can result in overestimation of the concentration of sample, under-seeding of the target amplification reaction, and low library yields.
We recommend using the Qubit dsDNA HS Assay Kit (Cat. No. Q32851) for quantifying the cfDNA samples.
We recommend using the MagMAX Cell-Free DNA Isolation Kit (Cat. No. A29319), which was included in-house during the Oncomine cfDNA workflow verification testing.
