No-Weigh 微量管处理:

What is the protocol for crosslinking IgG to Dynabeads from the Dynabeads Protein G Immunoprecipitation Kit (Cat. No. 10007D) using DSS (disuccinimidyl suberate)?

For crosslinking the antibody to Protein G Dynabeads, please refer to the crosslinking protocol with BS3 (bis(sulfosuccinimidyl)suberate) or Sulpha-DSS, which is an analogue of DSS. The only difference between these two molecules is that DSS is water insoluble and needs to be solubilized in DMSO before use. You can find the protocol at the following link.

How does a product with "No-Weigh" format reduce my concern over reagent stability?

The No-Weigh product format, where small amounts of reagent are pre-weighed and packaged separately, prevents the contamination and loss of reagent reactivity over time associated with repetitive opening and closing of a single vial. This format enables use of a fresh vial of reagent each time and eliminates the hassle of weighing out small amounts of reagent. Do you recommend crosslinking reagents for viral capsids (diameter 30 nm)?

Amine-to-amine crosslinkers are a good choice for your application since amino groups are usually abundant on the surface of proteins. Our most popular amine-reactive homobifunctional crosslinker that is water-soluble is BS3 (Cat. No. A39266). This has a moderate-length (11.4 A) linker that is good for linking adjacent proteins. If you need the ability to cleave the linker with sulfhydryl reducing agents I would suggest DTSSP (Cat. No. 21578). If you think you might need an extra-long crosslinker, then BS(PEG)5 (Cat. No. 21581) or BS(PEG)9 (Cat. No. 21582) are alternatives.

I would like to use BS3 (Cat. No. A39266) to crosslink a membrane-bound receptor to its ligand and analyze the resulting complex via Western Blotting. Will the complex remain intact when boiled and run in the presence of SDS?

The crosslinks formed by BS3 are stable to SDS/heat treatment as used for gels and blots.
