When using frozen pellets as a starting point, please consider the following guidelines:

The pellet should represent 150 mL of the specified culture.
Fully resuspend the pellet in 150 mL of 1X PBS or the parent media.
The resuspension can be achieved by either pipetting up and down or vigorous vortexing.
