Can I resuspend a frozen pellet for use with the KingFisher PlasmidPro Maxi Processor?

When using frozen pellets as a starting point, please consider the following guidelines:

  • The pellet should represent 150 mL of the specified culture.
  • Fully resuspend the pellet in 150 mL of 1X PBS or the parent media.
  • The resuspension can be achieved by either pipetting up and down or vigorous vortexing.

No. The KingFisher PlasmidPro Maxi Processor does not require any downtime between runs. The system does not employ any mechanical, optical, or electrical components that have a “wait” time to reset between purification runs. As soon as the used cartridge is removed from the instrument, a new cartridge can be inserted and run immediately.

What are the guidelines for growing E. coli for optimal yield and purity results when using the KingFisher PlasmidPro Maxi Processor, e.g., OD range, growth media, plasmid size?

  • Use overnight Luria-Bertani medium optimally grown for at least 12 hr but not greater than 16 hr. Grow sufficient culture volume to support 150 mL.
  • Use bacterial cultures with a measured cell density of approximately 1.5 - 6.0 absorbance at 600 nm (A600).
  • Use a high-copy or low-copy plasmid between 3 and 10 kb cultured in Luria-Bertani medium to obtain maximal yield and purity of plasmid DNA.

Note: Cell density or plasmid kb outside of the recommendation will result in reduced yield and purity.

The KingFisher PlasmidPro Maxi Processor is designed for fully automated, maxi-scale plasmid purification from cultures grown in Luria-Bertani medium or Terrific Broth. The instrument utilizes a self-contained cartridge pre-filled with the necessary reagents to process the culture by filtration and alkaline lysis, to minimize handling and preparation time. It uses a system of pumps and valves to move the sample and reagents in the cartridge to perform all the steps of the purification process in under 75 mins and provides endotoxin-free plasmid DNA resuspended in TE buffer, at the end of the procedure.
