No. The labeling is not retained upon fixation. Imaging of the samples should be done only on live cells. To limit photobleaching while viewing live cells, use ProLong Live Antifade Reagent, for live cell imaging (Cat No. P36974 and P36975).

Docetaxel has a slightly higher binding affinity than Taxol and exhibits longer retention time due to better uptake and slower efflux, but for use in labeling cultured cells, these differences should be minimal. Tubulin Tracker Deep Red is not acetylated. It is inherently cell permeable and is not modified further by any cellular enzymatic activity to fluoresce or bind to actin.

The use of Probenecid is optional. Its requirement is dependent upon the cell type. Some cell types actively efflux the reagent out of the cells within a short time, limiting detection, other cell types do not exhibit this activity or exhibit very limited activity.
