工作原理 用于将 DNA 引入哺乳动物细胞的磷酸钙转染方法的基础是形成磷酸钙 - DNA 沉淀。磷酸钙可促进 DNA 与细胞表面的结合。DNA 然后通过内吞作用进入细胞。该方法首先由 Graham 和 van der Ebb 开发,后来由 Wigler 进行了修改。该程序常用于转染多种用于瞬时表达或产生稳定转化体的细胞类型。将 DNA 直接与 CaCl2 浓缩溶液混合,然后逐滴加入磷酸盐缓冲液中以形成精细沉淀。加入 DNA-CaCl2 溶液的同时对磷酸盐缓冲液通气有助于确保形成尽可能精细的沉淀,这一点非常重要,因为聚集的 DNA 不会有效地粘附到或进入细胞。
