Neural stem cells are a unique population of cells capable of self-renewal and differentiation into various neural cell types, including neurons, astrocytes, and oligodendrocytes. NSCs are typically made in specific regions of the brain, such as the subventricular zone (SVZ) and the hippocampus, where they reside in a niche that supports their growth and differentiation. Culturing NSCs in vitro allows researchers to study their properties, differentiation pathways, and potential therapeutic applications in neurodegenerative diseases and brain injuries.
To culture NSCs, researchers can isolate them from neural tissue and maintain them in a specialized growth medium containing essential nutrients and growth factors like EGF and FGF-2, which promote self-renewal and proliferation. NSCs can also be generated from pluripotent stem cells (PSCs) in vitro through a process called neural induction. Conventional methods of neural induction from PSCs include embryoid body (EB) formation and isolation of neural rosettes or co-cultures with stromal cell lines. These methods have several disadvantages including a time-consuming protocol and variability in the quality of resulting NSCs. Today, more efficient methods leverage small molecules and signaling pathways in a monolayer culture.
Neural stem cells are important because of their ability to self-renew and their multipotent capacity to differentiate into neurons and glial cells, which have essential functions in the nervous system. NSCs are not only a valuable source of cells for neuroscience research but also have potential as a regenerative medicine to help treat neurodegenerative diseases and other neurological disorders.
