Traditional fermentation methods that use complex media can introduce risk and variability that slow process development and complicate technology transfer. Gibco Bacto CD Supreme media and feed offer a chemically defined alternative for microbial fermentation compatible with batch and fed-batch processing, available in flexible formats for process development through manufacturing. These formulations are designed to support the production of plasmid DNA and recombinant protein in Escherichia coli (E. coli).

Use Gibco Bacto CD Supreme FPM Plus with Bacto CD Supreme Feed (2X) to support high biomass, defined fed-batch control, and scalable plasmid DNA production in E. coli. In a fed-batch process we boosted pDNA to over 250 mg/L and showed consistent performance as process scaled from development studies to a 30 L Thermo Scientific HyPerforma Enhanced Single-Use Fermentor (e.S.U.F.).
Figure 1. NEB Stable cell density. Bacto CD Supreme FPM Plus with Bacto CD Supreme Feed supported up to 151% higher cell growth compared to TB with the lab chemically-defined, serum-free (CDSF) feed. Bacto CD Feed supported 129% higher cell growth relative to using the CDSF feed with Bacto CD FPM Plus. (n = 2–3 vessels per condition)
Figure 2. pAAV2-GFP ITR pDNA titer. Up to 120% higher average pAAV2-GFP-ITR* pDNA titers with Bacto CD Supreme FPM Plus and Bacto CD Supreme Feed. A 38% increase in pDNA was realized using Bacto CD Feed compared to using the CDSF Feed with Bacto CD FPM Plus. (n = 2–3 vessels per condition)
Figure 3. Cell density with scaling. Across scaling to 30 L, similar growth profiles and peak cell densities of ~90 to 120 OD were supported. (n = 2 per 250 mL vessel, n = 3 per 3 L vessel, and n = 1 per 30 L vessel)
Figure 4. pIVT production with scaling. pIVT production was shown to be comparable with scaling, ranging from 250 to 306 mg/L. (n = 2 per 250 mL vessel, n = 3 per 3 L vessel, and n = 1 per 30 L vessel)
For recombinant protein workflows, the same platform supports strong cell density and reproducible expression performance. In the current dataset, Bacto CD Supreme FPM Plus with Bacto CD Supreme Feed (2X) supported an average cell density increase of up to 74% and 55% compared with Terrific Broth paired with chemically defined serum-free feed or Bacto yeast extract, respectively, while supporting comparable enhanced green fluorescent protein (GFP) production.
Figure 5. Cell density. An average cell density increase of up to 74% and 55% using Bacto CD Supreme FPM Plus with Bacto CD Supreme feed compared to TB paired with CDSF or Bacto Yeast extract, respectively. Bacto CD Supreme FPM Plus with CDSF feed supported a 54% increase in cell density versus using TB with the CDSF feed. (n = 2 vessels per condition)
Figure 6. GFP productivity. Bacto CD Supreme FPM Plus with either CD feed supported comparable to enhanced green fluorescent protein (GFP) production compared to TB with either the yeast extract or CDSF feeds. (n = 2 vessels per condition)
Complete your upstream workflow by pairing Gibco Bacto CD Supreme media and feed with the Thermo Scientific HyPerforma Enhanced Single-Use Fermentor System. Designed specifically for microbial fermentation, the HyPerforma e.S.U.F. platform supports oxygen transfer, mixing, and temperature control for high-density microbial culture and is available in 30 L and 300 L formats to support process development and scale-up.
This pairing helps support a workflow solution spanning media selection, fed-batch optimization, and scalable single-use fermentation, while supporting faster turnaround, lower contamination potential, and reduced facility burden compared with traditional reusable systems.
Cell culture media support a range of upstream applications for vaccine development workflows. These formulations are designed to enable reliable performance across multiple production systems, helping teams maintain reproducibility from early development through scale-up.
Chemically defined media use fully specified ingredients and concentrations, which helps reduce the variability associated with hydrolysates or peptones and supports more reproducible microbial fermentation performance.
Gibco Bacto CD Supreme FPM is a chemically defined dehydrated powder medium designed for microbial fermentation and high-cell-density E. coli culture. Gibco Bacto CD Supreme FPM Plus extends the platform with broader strain compatibility, batch and fed-batch positioning, and both liquid and dry powder format options for plasmid DNA and recombinant protein workflows.
Use Gibco Bacto CD Supreme Feed (2X) for fed-batch microbial fermentation when you want to extend culture performance and increase productivity with a defined nutrient strategy that integrates with the Bacto CD Supreme media platform.
Yes. Gibco Bacto CD Supreme FPM Plus and Feed are positioned for both workflows, and the current draft data set includes performance claims for plasmid DNA production and recombinant protein expression.
Yes. The product pages describe the formulations as chemically defined and free from animal-origin materials or components.
Yes. The current draft cites consistent performance through a 30 L HyPerforma e.S.U.F., and the live fermentor systems page positions the HyPerforma e.S.U.F. as designed specifically for microbial fermentation in 30 L and 300 L formats.
Depending on the product, the portfolio is available in both liquid and dehydrated powder formats to support development and manufacturing workflows.
The portfolio supports filter sterilization or autoclave-compatible workflows, depending on the product and process needs.
Bacto CD Supreme media and feed are for use in microbial cell cultures to improve overall culture performance in batch or fed-batch processes. Before use, you need to add a carbon source to Bacto CD Supreme media and/or feed. We recommend glycerol or glucose (dextrose); however, the choice and concentration may vary based on your current laboratory process.
