Low fluorescence PVDF membranes

 

Low-fluorescence PVDF membranes are the preferred choice for fluorescent and near-infrared western blotting, minimizing autofluorescence to improve signal-to-noise ratios and detection sensitivity.

Low-fluorescence PVDF membranes are engineered to reduce autofluorescence across commonly used fluorescence channels, enabling more accurate detection and improved quantitative results.

 

Reduced autofluorescence across detection channels

 

Invitrogen low fluorescence PVDF membranes exhibit low auto-fluorescence across all fluorescent channels and result in reduced background compared to both nitrocellulose and standard PVDF membranes (Figure 1).

Consistent membrane structure for reliable performance

 

Invitrogen low fluorescence PVDF membranes feature a 0.3 µm pore size with high consistency and uniformity across the membrane surface (Figure 2), that contributes to consistent protein binding and improved reproducibility across experiments.

Exceptional performance at lower wavelength channels

 

Compared to other commercially available low fluorescence PVDF membranes, Invitrogen low fluorescence PVDF membranes offer excellent performance when imaging in lower wavelength channels (Figure 3). Reduced autofluorescence is especially important at lower wavelengths, where background signal can significantly impact detection sensitivity.

Unprocessed membranes were hydrated and imaged together in six different fluorescent channels on a compatible digital western blot imaging system using exposure times and contrasting consistent with standard western blot applications. Within each channel, contrasts and imaging time were consistent for each membrane. Membranes were hydrated in 100% methanol for 3 minutes and washed in diH2O for 5 minutes on an orbital shaker.

Improved signal-to-noise in multiplex experiments

 

Reduced background fluorescence enables detection of more protein targets per blot and supports higher-quality, publication-ready data.

  • Higher signal-to-noise ratio = clearer, more distinguishable bands (Figure 4)
  • Improved detection sensitivity = better limits of detection for low-abundance proteins
  • Enhanced multiplexing performance = more reliable results across multiple targets, especially in low-wavelength channels

Figure 4. Invitrogen low fluorescence PVDF achieves higher signal-to-noise ratio in western blotting compared to other commercially available low fluorescence PVDF membranes. Four lysates were 2-fold serially diluted from 20 µg–2.5 µg, loaded and electrophoresed on Tris-Glycine gels, and transferred to three different membranes. Blocking was completed for 1 hour at room temperature, followed by overnight 4°C incubation with primary antibodies against Hsp90, Calreticulin, beta-Actin, and p23. Secondary antibodies (GAR-800, GAM-647, and GAC-488) in TBST were applied for 1 hour at room temperature. Blots were imaged and contrasted on the iBright FL1500 Imaging System under identical conditions.

Together, these results demonstrate how low-fluorescence PVDF membranes improve sensitivity, reduce background, and enable more reliable fluorescent western blot workflows.


Filter papers for western blotting

Filter papers support protein transfer by maintaining even contact and consistent buffer flow. Western blotting filter papers are pre-cut cotton sheets for wet or semi-dry, passive or electrophoretic transfer of proteins. High-quality western blotting filter papers are available as standard-thickness and extra-thick sheets that are pre-cut for convenience with mini gels and midi gels.

Frequently asked questions

Nitrocellulose and PVDF membranes both produce high-quality western blot results, with nitrocellulose commonly preferred for routine chemiluminescent, colorimetric, and fluorescent detection methods, while PVDF membranes help provide increased durability for chemiluminescent applications that require stripping and reprobing.

Low-fluorescence PVDF membranes are engineered to minimize autofluorescence across fluorescent channels, enabling improved signal-to-noise ratio, longer exposure times, enhanced multiplexing, and more accurate quantitative analysis, making them ideal for fluorescent western blotting and total protein normalization workflows.

Low-fluorescence PVDF membranes perform well for both fluorescent and chemiluminescent detection, offering high protein-binding capacity, durability, and low background fluorescence, making them a versatile option for laboratories that use multiple detection methods.

Membrane pore size generally has modest impact for most proteins, but smaller pore sizes improve retention of low-molecular-weight or low-abundance proteins. Ultimately, the optimal choice may depend on the membrane type and manufacturer, and require optimization based on the target protein.

Western blot membranes can be dried after transfer and stored for later detection, with PVDF requiring reactivation in methanol or ethanol before blocking and antibody incubation steps; drying may strengthen protein binding and support stripping and reprobing, but may increase background in some applications and should be verified.

Nitrocellulose membranes can be striped and reprobed, but they are more fragile and typically support only one or two stripping cycles. For repeated stripping, PVDF membranes are preferred due to their greater mechanical durability.
